I have a windows7-64bit OS Virtual Machine running on Fusion5 that won't launch the MP. I have all the right versions based on what I could see for requirements.
The error I am getting is as follows:

Qground or Mavlink has a problem with Win7 now it seems ,cannot run unless Net Framework 4.6.1 is loaded. Win7 does not accept it

to me it sounds like mavcmd.xml is currupt. please try uninstalling and reinstalling.
Had the same issue, but with version 4.
All I do is make sure I run the program as 'administrator' to do any installs or updates.
So when I run it normally and I get 'update now' , I just quit and then right click the app and select 'run as administrator'.
You may need to do teh install from scratch as 'administrator' as well to get past this.
